Abstract: Yeast alcohol dehydrogenase is an enzyme which is extremely interesting for industrial immobilization aimed at regeneration of coenzymes, fine organic synthesis, and analytical determination of alcohol in various media. The most important use in practice is preparation of immobilized ADH attached to inorganic supports which are characterized by good mechanical properties and high stability in aqueous solutions. Therefore, such systems can be used in a column regime with perfected hydrodynamic properties compared with enzymes on organic resines and in gels. However, all known methods of immobilization of ADH on inorganic do not allow preparation of enzymes with high stability which is one of the most important operation characteristics. We have attempted to obtain a stable and active immobilized yeast ADH by selecting an optimal pore structure of the support and optimal acid-base characteristics at its surface. For this purpose, we used alumina, a highly available and inexpensive support with easily controlled porosity. In order to change the acidity of the surface, the surface was successively carbonized via the catalytic cracking of hydrocarbons or modified by baselike substances capable of adsorbing on acid sites. As a result, a stable adsorbed ADH has been obtained. A further increase in stability can be reached by treating the enzyme with condensating agents such as glutaric dialdehyde and carbodiimide. The proposed method opens up new possibilities for practical utilization of yeast alcohol dehydrogenase.
