Abstract:
The dependence of change in catalytic properties of cytochrome P-450-containing microsomal monooxygenases on the structure of substrate and inducer molecules was studied. Benzene, cyclohexane, and tetralin were used as substrates and tetralin, phenobarbital, and 3-methylcholanthrene as inducers. The rate of hydroxylation of the substrates increased in the order benzene < cyclohexane < tetralin. There was a qualitative correspondence between the transition of the low-spin form of cytochrome P-450 to the high-spin form and the rate of hydroxylation. Induction by phenobarbital did not change the rate of hydroxylation of benzene, but sharply increased the rates for the other substrates. Induction with 3-methylcholanthrene on the whole decreased the rates of reaction. Estimations of the amount of increase in cytochrome P-450 concentration or the relative amount of low-spin form were not sufficient to account for the observed effects. Induction by tetralin significantly increased the rate of hydroxylation of tetralin at the α- and β-positions, although the effect was somewhat less than that with phenobarbital, and did not change the amount of cytochrome P-450. However, an increased binding of tetralin to induced cytochrome P-450 was observed compared to control. It is proposed that the change in catalytic activity of enzymes under the effect of inducers is due to the change in conformational state of the enzyme on binding of the substrate-inducer with subsequent fixation of this state on attachment to the membrane.
